The field of Explainable AI (XAI) aims to explain the decisions made by machine learning models. Recently, there have been calls for more rigorous and theoretically grounded approaches to explainability. In my thesis, I respond to this call by investigating the properties of explainability methods theoretically and empirically. As an introduction, I provide a brief overview of the history of XAI. The first contribution is a novel theoretically motivated attribution method that estimates the importance of each input feature in bits per pixel, an absolute frame of reference. The method is evaluated against 11 baselines in several benchmarks. In my next publication, the limitations of modified backward propagation methods are examined. It is found that many methods fail the weight-randomization sanity check, and the reasons for these failures are analyzed in detail. In a follow-up publication, the limitations of one particular method, Deep Taylor Decomposition (DTD), are further analyzed. DTD has been cited as the theoretical basis for many other attribution methods. However, it is found to be either under-constrained or reduced to the simpler gradient x input method. In the next contribution, a user study design is presented to evaluate the helpfulness of XAI to users in practice. In the user study, only a partial improvement is observed when users are working with explanation methods compared to a baseline method. As a final contribution, a simple and explainable model of the k-nearest neighbors regression is proposed. We integrate higher-order information into this classical model and show that it outperforms the classical k-nearest neighbors, while still maintaining much of the simplicity and explainability of the original model. In conclusion, this thesis contributes to the field of XAI by exploring explainability methods, identifying their limitations, and suggesting novel, theoretically motivated approaches. My work seeks to improve the performance and interpretability of explainable models toward more transparent, reliable, and comprehensible machine learning systems.
Weniger anzeigenDie vorliegende Dissertationsschrift widmet sich dem Autoritätsproblem in Uwe Tellkamps Wenderoman Der Turm. Geschichte aus einem versunkenen Land (2008). Tellkamps Roman stellt die DDR nicht als einen Staat dar, der plötzlich zusammenbricht, sondern als einen Staat, der langsam untergeht. Der Untergang des Staates ist dabei im Roman auf grundlegende Weise mit der Erosion staatlicher Autorität verbunden. Auffällig ist hierbei, dass der Roman den Untergang der staatlichen Autorität mit dem Untergang der väterlichen Autorität in der späten DDR parallelisiert. Anhand des exponierten Korrespondenzverhältnisses beider Instanzen arbeitet die Studie die Kritikwürdigkeit der Autorität der späten DDR-Zeit im Roman heraus. Im Rahmen einer Analyse der im Roman verhandelten Autoritätsstrukturen kann in Kapitel 2 gezeigt werden, dass die kulturell konservative Lebensführung sowie die opportunistisch-adaptive politische Einstellung der Romanfiguren Reaktionen auf den Zusammenbruch der staatlichen Autorität darstellen. In Kapitel 3 macht die Untersuchung deutlich, dass die DDR zwar einen hypertropher Erziehungsraum bildet, dass persönliche Bildung hier jedoch letztlich unmöglich ist. Es wird dabei die These vertreten, dass Tellkamps Roman in der Form des Antibildungsromans nicht nur die Erziehungsversuche des Staats, sondern auch die Erziehung durch die Vätergeneration kritisiert. In Kapitel 4 wird gezeigt, dass die Erosion der staatlichen und väterlichen Autorität mit ihren jeweiligen moralischen Defiziten zusammenhängt und dass die Gewalt des Staates und des Vaters als Substitut ihrer geschwundenen Autorität nicht auf Dauer aufrechterhalten werden kann. In Kapitel 5 wird argumentiert, dass der Roman einerseits die Beschränkung speziell der männlichen Autorität des Staates und des Vaters aufzeigt und dass er andererseits ein alternatives Autoritätsmodell präsentiert, das horizontal und demokratisch funktioniert. In Kapitel 6 wird der Nachweis geführt, dass das Problem der Autorität in Der Turm eng mit einer Krise der Kommunikation verknüpft ist. Die Arbeit untersucht in Kapitel 7 das geschwächte Zugehörigkeitsgefühl der Ostdeutschen zu ihrem Staat, der in der späten Phase der DDR seine Autorität bereits weitgehend eingebüßt hatte. Die Arbeit macht deutlich, dass der Zusammenbruch der DDR für viele Ostdeutsche nicht notwendigerweise ein ungebrochen positives Erlebnis war, sondern mitunter auch zu Orientierungslosigkeit oder zu ambivalenten Gefühlen führte. Abschließend wird argumentiert, dass die Betrachtung der späten DDR-Gesellschaft, die in Tellkamps Roman als Autoritätsvakuum erscheint, einen möglichen Ausgangspunkt für ein umfassenderes Verständnis der Lebensrealität der ehemaligen DDR(-Bürger) und damit auch der Ursachen für die bis heute ungelösten Probleme nach der Wiedervereinigung bildet.
Weniger anzeigenThis dissertation deals with the economy of Sonnenallee, a boulevard in Berlin’s district Neukölln which is famous for its many small- and micro-sized-businesses run by immigrants. Whereas until the early 2000s, the image of Neukölln dominated public debates as an example for the failed state of the multicultural society, in recent years the Berlin senate concentrated on a location marketing for this district and especially Sonnenallee as a symbol for diversity, inclusion and creativity, contributing fundamentally to the location’s rising popularity. Known around the city as “the street of Arabs”, today Sonnenallee attracts many customers as well as potential investors, creating an intense competition among entrepreneurs and workers alike and resulting in highly precarious working conditions. Based on field work over the course of five years, what becomes obvious in this research is the significant control private property owners have in Sonnenallee. Due to the scarcity principle, private property owners can charge monopoly rents which lead to eminent wealth disparities among entrepreneurs who are sometimes sub-contractors rather than company owners. By conducting a historical analysis, this thesis explores what kind of economic opportunities the street offers in a given period of time for immigrants from the Middle East and highlights the variety of forms that self-employment can take on.
Weniger anzeigenThe field of supramolecular chemistry is conceptually very broad, encompassing diverse classes of compounds with different functions and uses. Robust characterisation of the systems is one of the most important steps in building the knowledge necessary for the further development and eventual use in numerous potential applications. Establishing additional methods to characterize supramolecular complexes would be beneficial as it is currently dominated by NMR and X-ray crystallography. Further use of mass spectrometry (MS) would be useful for supramolecular chemistry as it is highly versatile and able to provide detailed structural information. Throughout this thesis, MS was used to structurally characterise a diverse range of supramolecular systems including several different types of host-guest systems, a photoswitchable pseudorotaxane, a light driven molecular motor, and a bioderived rotaxane. Structural insights into the different complexes could be obtained by using various combinations of the tandem MS techniques collision induced dissociation (CID), ion-mobility mass spectrometry (IMS), and hydrogen deuterium exchange. The general results show the ability of MS to build on other characterisation methods to gain further insights into the system under investigation. More specifically, it shows how MS can be used to follow dynamic processes of photoswitchable systems. For this, IMS is particularly useful as the different conformations can be separated and then assigned based on their collision cross section values. In addition, IMS can be combined with CID to investigate thermal relaxation processes. There are also situations where the complexes may only be observed via MS. Here guest binding was seen via MS that was not observed via other techniques and the binding mode elucidated via IMS and CID measurements. The results show how the ionisation process can be used to generate complexes that will not form otherwise. This is particularly prevalent for host systems binding charged guests as the charged droplets enable interaction between the naked ion and the host. Overall, the results show that MS is a powerful tool in the structural characterisation of supramolecular systems and can provide information not possible by other methods.
Weniger anzeigenThis dissertation examines contesting practices of visual activism in the social movement environment of today that is largely shaped by digitality. Despite the substantial amount of research on the role of the Internet and social media in the emergence and trajectory of social movements, little is known about how mobilisations with incompatible political goals influence each other on digital platforms and in visual terms in particular. Addressing the ways in which online visuals mobilise people in contemporary social movements across political divides, this study shows that political imageries developed on different sides of a social conflict can be remarkably similar in form and content despite their diverging viewpoints. At the same time, the study argues that the possibilities of developing a perennial character of images online is a crucial factor in configuring the interaction between political divides and that leads to the rise of common and anonymous people as icons that mark social movements in digital societies.
Accordingly, the dissertation offers a comparative analysis of still images collected in relation to Turkey’s 2013 Gezi Movement and 2016 Anti-Coup Resistance, two popular mobilisations that had diverse political goals. In order to address both the images produced and disseminated online during the peak times of the mobilizations as well as the extended low-level activism that continued beyond these peaks, data collection covers a five-year period between 2013 and 2018. Photographs and illustrations found on social media platforms, blogs, and digital news portals are analysed through grounded visual analysis, an original model combining the qualitative approaches of grounded theory with established visual research methods. Following a sociological methodology but at the same time drawing from Art History, Cultural Studies, and Media and Communications, this dissertation also makes an interdisciplinary methodological contribution to the study of social movements.
Furthermore, in order to interpret the findings, the project combines framing, a much- used theoretical approach in social movements research borrowed from Goffman, with everybody, a concept that can be found in the work of de Certeau and, to some extent, in the growing literature after Deleuze and Guattari’s work on affect. The results of the study show that photographs establish similar visual frames, yet with certain differences, across political divides, while illustrations contribute to these frames in diverse ways. Some of the illustrations consolidate them, and others, particularly those that remediate photographs, promote certain figures that sharpen, deepen, and layer the initial frames, paving the way for the iconisation of images.
Ultimately, the study highlights the convergence and struggle over the symbolic dimension of social movements with incompatible political goals and situates the mobilising role of ordinary figures as the main focus of visual activism today.
Weniger anzeigenUsing variational principles, we investigate elastic rod structures under external loads that are clamped at one end and free at the other. Their stability properties are analyzed using second order conditions by generalizing the Jacobi theory of conjugate points. The notion of the index, which quantifies the dimension of the subspace of variations over which the second variation is negative, is also generalized to this class of problems. The variational structure of the parameter- dependent calculus of variations problems can be exploited to detect the changes in the index at the folds as the parameter is varied, with the assistance of distinguished bifurcation diagrams. We generalize these plots to cover our current case with fixed-free ends. Furthermore, we extend the investigation to the problems with discontinuous integrands by generalizing the concept of conjugate points, index, and distinguished bifurcation diagrams to them. For this purpose, second-order matching conditions are derived at the points of discontinuity. These techniques are developed with the aim of employing them in soft robotic applications, a field that is increasingly gaining popularity. Applications such as Concentric Tube Continuum Robots (CTCRs) employ intrinsically curved rods to generate flexible mechanisms. We emphasize the impact of intrinsic curvature, which is often a source of complex mechanics, on the equilibria of elastic rods and their stability. The interplay between geometric non-linearities, external load, and intrinsic curvature leads to intriguing and complex behavior such as snap-back instability. We study the influence of the parameters, such as intrinsic curvature, length, tip load, and lever arm of the load on this behavior. This study aids in their efficient utilization in practical applications. We extend this investigation to CTCRs, which resemble an intrinsically curved elastic rod with slightly different physics. These robots consist of multiple sections, and their properties change abruptly at the boundary of each section. This research has the potential to advance the design and control of robots for tackling more complex tasks. Finally, an open-loop gradient-based navigation is devised to model the robot maneuver using optimal control techniques. Through this approach, various tasks can be modelled in terms of objective functions that are subsequently optimized. We consider optimal control of CTCRs parameterized over pseudo-time, primarily focusing on minimizing the robot’s working volume during its motion. A numerical strategy to implement this optimization task is also discussed. This optimal control-based methodology can be adapted to any backbone-based continuum robots.
Weniger anzeigenDie synergistische Zytotoxizität zwischen Ribosomen-inaktivierenden Proteinen und Triterpensaponinen beruht auf dem Triterpensaponin-vermittelten endosomal escape von Typ I RIPs. Typ I RIPs wirken zytotoxisch, indem sie einen spezifischen Adeninrest von der rRNA abspalten. Die Abspaltung hat das Erliegen der Proteinsynthese und den Zelltod zur Folge. Damit Typ I RIPs ihre Toxizität ausüben können, müssen sie aus den Endo- und Lysosomen freigesetzt werden, andernfalls werden sie proteolytisch abgebaut. Spezifische Triterpensaponine bewirken dabei, dass Typ I RIPs dem endolysosomalen Abbau entkommen, indem sie in das Zytosol freigesetzt werden. Bisherige Untersuchungen deuteten darauf hin, dass Triterpensaponine pH-abhängig an Typ I RIPs binden und dadurch die endolysosomale Freisetzung induzieren. Die essenzielle Glucuronsäure an C-3 der Triterpensaponine, negativ geladen in Endo- und Lysosomen, könnte dabei mit basischen Aminosäuren der Typ I RIPs, positiv geladen in Endo- und Lysosomen, interagieren. Voraussetzung für die weiteren Untersuchungen dieser Arbeit war das Erbringen des Nachweises der Bindung von SO1861 und HisDianthin, der bisher nur am Beispiel von HisSaporin-3 erbracht wurde. Massenspektrometrische Aufnahmen unter nativen Bedingungen zeigten bei pH 6,24 für SO1861 und HisDianthin eine Assoziation im stöchiometrischen Verhältnis von 1:1. Der Einfluss basischer Aminosäuren auf die bestätigte Interaktion zwischen SO1861 und HisDianthin wurde an 19 HisDianthin-Mutanten, bei denen Lysin- und Arginin-Reste durch Alanin ersetzt wurden, anhand ihrer synergistischen Zytotoxizität untersucht. Für die ausgewählten Lysin-Reste konnte keine Beteiligung an der synergistischen Zytotoxizität nachgewiesen werden. Einzig die synergistische Zytotoxizität von HisDianthin Arg24Ala war im Vergleich zu dem nativen HisDianthin reduziert. Die aufgetretene Reduktion der Zytotoxizität war nicht zelllinienabhängig. Allerdings war sie auch ohne Anwesenheit von SO1861 reduziert, bei gleichzeitig unbeeinträchtigter N-Glykosidaseaktivität. Die Markierung von HisDianthin Arg24Ala und HisDianthin mit Fluoreszenzfarbtsoff erlaubte via Durchflusszytometrie, die verminderte Zytotoxizität auf eine verringerte Endozytose zurückzuführen. Im Rahmen dieser Arbeit konnte somit gezeigt werden, dass die untersuchten Arginin- und Lysin-Reste der Typ I RIPs nicht hauptverantwortlich für die Interaktion mit SO1861 zu sein scheinen. Um aufzuklären, auf welcher Interaktion die Bindung zwischen Typ I RIPs und Triterpensaponinen, die vermutlich dem endosomal escape zu Grunde liegt, beruht, müssen weitere Experimente durchgeführt werden. Um die Funktion von Arg24 an weiteren Typ I RIPs validieren zu können, wurden aus den Samen der Gypsophila vaccaria (L.) Sm. die RIP-Isoformen Sapovaccarin-S1 und -S2 isoliert und sequenziert. Das Isoformengemisch wies eine für Typ I RIPs typische Proteinsequenz auf. Die N-Glykosidaseaktivität und die Thermostabilität waren mit denen von isolierten Typ I RIPs vergleichbar. Die Untersuchung der Funktion der Aminosäure Arg24 an den Typ I RIPs HisSapovaccarin-S1 und HisAgrostin bestätigte, dass auch hier eine Reduktion der Zytotoxizität zu verzeichnen war. Die durchgeführten Untersuchungen deuten darauf hin, dass die Funktion von Arginin an Position 24, die am HisDianthin aufgezeigt werden konnte, sich auch auf die Endozytose von HisAgrostin und HisSapovaccarin-S1, wenn nicht sogar auf Typ I RIPs im Allgemeinen übertragen lässt.
Weniger anzeigenThe respiration rate (RR) is a suitable parameter for assessing the health status of cattle, as it is very sensitive for detecting stress or pain, and thus shows an indisposition in animals at an early stage. Furthermore, it is a suitable parameter for the early detection of heat stress, which becomes increasingly important due to climate change. In order to determine the proportion of heat emitted through respiration, an additional measurement of the tidal volume (Vt) is necessary. The measurement of Vt is also suitable to assess the performance of the lungs and allows conclusions about respiratory and lung diseases. The gold standard methods for both parameters are time consuming and can also influence the behavior of the cow. Counting the cow's flank movements is often used as the method of choice for recording RR. However, different methods are found in literature regarding duration and type of counting. While in some studies, the RR was counted for 5 or 10 breaths and then extrapolated to 60 s, other authors counted the RR for 15, 30 or 60 s. In order to determine which method is the most precise, Publication “A” compared the 5 methods mentioned above. The reference method used was an RR sensor that reliably measures the RR based on the pressure difference between inhalation and exhalation. In Publication "A", it was shown that counting the RR for 60 s is the most accurate method and that as the RR increases, the inaccuracy of the other methods continues to increase compared to counting for 60 s. Since a facemask has to be fitted around mouth and nostrils to measure Vt, the second fundamental parameter in lung function diagnostics, by a pneumotachograph, the animals have to be restrained during the entire experiment and since the behavior is severely restricted by the mask, only short measurement periods are possible. Therefore, the driving force for Publication "B" was to investigate whether it is possible to derive a Vt equivalent from the measured pressure of the RR sensor. A successful outcome would greatly simplify the measurement method, as the animals only have to be fixed for measurements with the RR sensor and can then move freely in the barn, making longer measurements possible. However, it was shown in Publication "B" that technical adaptations to the RR sensor are necessary in order to be able to reliably determine the Vt equivalent. Furthermore, there are approaches in human medicine to determine a relative Vt using infrared thermography, which can also be used to calculate the RR based on the temperature difference of the inhaled and exhaled air. The trend in livestock research is similar – sensors attached to the animal are being replaced by contactless systems for data acquisition. The main advantages of this alternative is that one sensor can record multiple animals and no animal fixation is needed to attach the sensors, which also reduces the risk of injury to humans. For this reason, we are currently testing infrared as well as depth cameras at ATB in cooperation with the University of Hildesheim, the Dida Data Science GmbH, and LVAT as part of the KAMI project (Artificial intelligence for measuring respiration in dairy cows). The aim of future studies is to use artificial intelligence and imaging techniques to automatically record the RR and integrate this parameter into existing herd management systems.
Weniger anzeigenIm Rahmen dieser Dissertation wurde ein breites Spektrum an Nage- und Beuteltierproben analysiert. Dies führte zu vielen Interessanten Entdeckungen, wie etwa die große Prävalenz an Picobirnaviren oder die Detektion von mindestens zwei, bis dato unbekannten, neuen Genera aus der Familie Picornaviridae. Zudem gelang erstmaligen Nachweis eines Deltavirus aus einem Säugetier abseits vom Menschen, sowie dessen molekularbiologische Charakterisierung und Unterschied im Replikationsverhalten gegenüber Hepatitis Deltaviren. Darüber hinaus brachte die Detektion und bioinformatische Analyse der Hepaciviren einen enorm großen Wissensgewinn. Darüber hinaus konnte die Theorie, die hinter dem ökologischen „Verdünnungseffekt“, nämlich die Abhängigkeit von Speziesdiversität zu Prävalenz, klar bestätigt werden. All diese Untersuchungen verschiedener Proben um den Gatún See in Panama zeigten viele, menschengemachte Einflüsse auf die ansässigen Viruspopulationen auf. Durch den Bau des Panama Kanals wurden weite Teile des Urwalds überflutet, woraufhin der Gatún See entstand. Zusätzlich zu diesem initialen Terraforming, kam es durch den wirtschaftlichen Aufschwung zu einer Ausbreitung urbaner Flächen. All dies führte dazu, dass der Mensch mehr Kontakt zu diversifizierenden, potenziell zoonotischen, sowie bislang noch unbekannten Viruspopulationen hat. Und wie uns die jüngste Zeit zeigt, ist die Gefahr einer Pandemie mit zoonotischen Ursprung allgegenwärtig und sollte nicht unterschätzt werden. Aus diesem Grund ist es wichtiger denn je die Zusammenhänge zwischen verschiedenen Einflussfaktoren zu verstehen und eine Ausbreitung beziehungsweise das Erscheinen neuer zoonotischer Erreger vorhersagen zu können. Die aktuelle Pandemie zeigt wie relevant das Wissen um die Auswirkungen dieser Faktoren auf Pathogenität und Infektiosität (bzw. Infektionsrisiko) ist. Die Ergebnisse dieser Arbeit sollen dazu dienen, wie im Fall der Deltaviren, den bislang unbekannten Ursprung dieser Familie Deltaviren zu ergründen. All diese Grundlagen können in weiterführenden Studien nicht nur helfen, Pathogen-Wirts Interaktionen besser zu verstehen, sondern auch mögliche medizinische und ökologische Relevanzen für den Menschen abzuschätzen.
Weniger anzeigenThe empirical studies assembled in this dissertation investigate pretax income inequality along three fault lines in Germany: inequality between different groups along the German income distribution, regional inequality between East and West German residents and gender earnings inequality. The last chapter turns to feasible policy solutions.
In Chapter 1, “Distributional National Accounts (DINA) for Germany, 1992-2016”, my co-authors, Stefan Bach and Charlotte Bartels, and I investigate inequality dynamics between different income groups – from the bottom to the very top – of the German income distribution from 1992 to 2016. Our analysis underlines the importance of firm structures for the level of top income concentration. The top 0.1% receives approx. 6% of national income, which is comparable to the United States and twice as high as in France. Due to Germany’s country-specific predominance of closely held partnerships, highly concentrated business profits can translate more directly into personal business incomes.
Chapter 2 “When Capitalism Takes over Socialism: The Lasting Economic Divide Between East and West Germany” turns to regional inequality. My co-author, Charlotte Bartels, and I estimate these differences along the regional income and wealth distributions from 1992 to 2016 and explore possible causes of the lasting divide. We find that East German residents still earn and own a fraction of their West German counterparts. This gap widens towards the top of the distribution, predominantly due to lower partnership and corporate incomes among East Germans.
Chapter 3, “The Long Way to Gender Equality: Gender Pay Differences in Germany, 1871-2021”, highlights another persistent fault line in German society: Gender earnings inequality. I provide the first long-run time series of the gender earnings ratio for the full-time employed workforce from 1871 to 2021, discuss possible drivers of the observed dynamics and compare the German path with the U.S. and Swedish cases. In Germany, the gender earnings ratio increased from about 46% in 1913 to 58% in 1937. Similar increases were visible in Sweden, and the U.S. Germany’s focus on on-the-job vocational training may have slowed educational convergence compared to the United States. Germany’s similar earnings ratio increase is better explained by women’s migration from low-paid agriculture to higher-paid white-collar jobs.
Chapter 4, “Revenue Effects of the Global Minimum Tax under Pillar Two”, turns to adequate policy action. Increased possibilities to shift profits to low-tax jurisdictions have raised fairness concerns for national tax systems and decreased national tax revenues. My co-authors, Mona Baraké, Paul-Emmanuel Chouc and Gabriel Zucman, and I explore the revenue potential of a global minimum tax of 15% on corporate profits. This policy was agreed upon among 137 countries and jurisdictions under the umbrella of the OECD in October 2021. We simulate the revenue effects of the global minimum tax. Our analysis accentuates the significance of policy design: We find that the distribution of revenues depends on which country has the priority to collect the top-up tax.
Weniger anzeigenIntrinsically disordered regions (IDRs) in proteins have been linked to many crucial functions, including mediating protein-protein interactions (PPIs), despite lacking a single invariant three-dimensional structure. This growing recognition has led to an increased demand for computational studies that focus on the amino acid sequences corresponding to proteins to identify crucial sequence characteristics in IDRs and their connections to diverse cellular functions. In the first part of this thesis,we have put forward two statistical methods to identify sequence features responsible for IDR functions. We introduce a statistical approach for quantifying the periodicity of aromatic residues in the human proteome by modeling their occurrence using a Poisson process. Next, we introduce another statistical analysis of IDR sequences to identify co-occurring amino acid groups in transcription factors (TFs) that co-bind to enhancer elements. In the second part of the thesis, our focus shifts to predicting PPIs using only protein sequences. In this thesis, we present a novel method to address PPI prediction challenge using IDR sequences. We encountered challenges while developing a PPI prediction model because our task essentially involves making predictions based on pairs of input data. In this regard, we present two distinct machine learning algorithms to address two different types of PPI prediction problems, namely, asymmetric and symmetric problems. For the asymmetric problem, where one of the proteins has already been included in the classifier, we develop a method to predict disordered protein partners of the known proteins in our dataset. On the other hand, for the symmetric problem, we implement another approach to predict entirely novel PPIs. Furthermore, we explore whether IDR amino acid sequences outperform other sequence components, including entire sequences and non-IDR regions, in predicting PPIs. Our findings led us to the conclusion that disordered regions are particularly valuable in predicting interactions between intrinsically disordered proteins. In summary, this thesis provides insights into dealing with paired nature datasets when developing machine learning models for PPI prediction and demonstrates how statistical approaches can be used to investigate IDR sequences for feature identification and predict PPIs based on IDR sequences.

Fragestellungen bezüglich ihrer Herkunft, formalen Entwicklung, Verbreitung, Herstellung, Überlieferung und Funktion wurden erörtert, um ihre Stelle im bronzezeitlichen Kulturinventar zu rekonstruieren. Als Interpretationsraum der Untersuchungen diente das Karpatenbecken, die Analysen beruhen in erster Linie auf dem Sichelmaterial des Gebietes von Ungarn, dessen Funde im Rahmen der Dissertation in einer katalogisierten Form dargestellt sind. 4435 Bronzesicheln von 432 Fundstellen wurden dokumentiert, beschrieben und abgebildet. Die Objekte umfassen die Zeitspanne von ungefähr 1000 Jahren, die Perioden BzA2–HaC.
Weniger anzeigenDas Monitoring der Mengen- und Spurenelementversorgung sollte in die Bestandsbetreuung der Milchviehherden integriert werden. Unspezifische Krankheitssymptome wie reduzierte Fruchtbarkeit, Wachstums- und Leistungsdepressionen können die Folge von Dysbalancen im Mineralstoffhaushalt sein. Jedoch sind diese immer nur Verdachtsdiagnosen, wenn keine geeignete Diagnostik eingeleitet wird. Dafür ist die Wahl des richtigen Probemediums wesentlich, um eine möglichst genaue Aussage zum Versorgungsstatus tätigen zu können.
Zielsetzung dieser Arbeit ist es, Leber als Untersuchungsmedium zur Bewertung der Mengen- und Spurenelementversorgung beim Milchrind zu untersuchen und zu beurteilen. Hierfür wird die hepatische Konzentration mit denen in anderen Medien verglichen. Anschließend erfolgt die Beurteilung der Eignung der unterschiedlichen Substrate für das jeweilige Element zur Aussage der Versorgungslage und der untersuchten Gruppen.
Zu diesem Zweck wurden über einen Zeitraum von zwei Jahren Proben in Beständen in Brandenburg, Sachsen, Sachsen-Anhalt, Thüringen und Mecklenburg-Vorpommern gesammelt. Das Probenmaterial bestand aus Blut-, Kot-, Leber-, Haar- und Futterproben. Der Datenpool umfasst 355 Einzel- und 30 Bestandsproben von klinisch gesunden Holstein-Friesian Kühen. Die Probenwerte der Medien Kot, Harn, Vollblut, Futtermittel und Haare setzen sich aus gepoolten Bestandsproben von je 10 Probanden zusammen. Es erfolgte eine Futtermittelanalyse für die TMR und die Konzentrationen der Mengenelemente Kalzium, Magnesium, Schwefel. Die Spurenelemente Kupfer, Zink, Selen, Eisen, Mangan und Kobalt wurden mittels ICP-OES bzw. ICP-MS bestimmt.
Für die Elemente Magnesium, Kupfer, Zink, Selen, Eisen, Mangan und Kobalt unterscheidet sich die Gruppe Kälber signifikant von den untersuchten Gruppen laktierende und nicht laktierende Kühe. Dies ist von besonderer Bedeutung, da bisher wenig Literatur zur Beurteilung der Konzentrationen bei Kälbern vorliegt. Für die Elemente Kalzium, Magnesium und Schwefel kann der Gehalt in der Leber nur bedingt zur Beurteilung der metabolischen Versorgungslage genutzt werden, so dass die Leberbiopsie keinen Mehrwert bringt. Eine zuverlässigere Interpretation kann durch die Auswertung und Kombination mit weiteren Substraten oder der Blutwerte erzielt werden. Für Kupfer ist die Leberbiopsie die Methode der Wahl zur Beurteilung der metabolischen Versorgungslage, sollte jedoch durch die Analyse von TMR und Kot für die nutritive Versorgungslage ergänzt werden, um die Antagonisten zu berücksichtigen. Zink zeigt eine sehr hohe Leberkonzentration, vor allem beim Kalb. Deshalb stellt die Biopsie das beste Diagnostikum zur mittelfristigen metabolischen Versorgung dar.
Für Selen ist die Leber ein ähnlich sensitives Medium wie Blut. Im Rahmen der Biopsie kann durch die Messung der Konzentration eine gute Aussage zur momentanen Versorgungslage getätigt werden, allerdings bringt sie keinen Mehrwert. Eisen kann vor allem bei Kälbern in hohen Konzentrationen in der Leber gemessen werden. Beim adulten Rind ist die Biopsie ein Diagnostikum, wenn die TMR einen zu niedrigen Eisengehalt aufweist. Das Ergebnis der Eisenkonzentration im Leberbioptat wird durch den eigenen Blutgehalt verfälscht, so dass eine Aussage unklar ist und weiterer Untersuchungen bedarf. Für das Medium Mangan ist die Leberbiopsie das Diagnostikum der Wahl, vor allem wenn es um Intoxikationen geht. Jedoch kann wegen der geringen verfügbaren Daten die Beurteilung der Lebermangankonzentration nur eingeschränkt bewertet werden und bedarf weiterer Analysen. Das Lebergewebe spiegelt das genaueste Ergebnis der Kobaltversorgung wider. In diesen Proben konnten höhere Konzentrationen als in den anderen untersuchten Medien gemessen werden, was eine Bewertung der metabolischen Versorgungslage verbessert.
Es sind weitere Untersuchungen zu empfehlen, um die hier ermittelten Werte zu untermauern. Dies stellt dann die Basis für die Entwicklung eines Überwachsungsprofils der metabolischen Versorgungslage beim juvenilen und adulten Rind über die Analyse von Leberbiopsieproben dar.
In dieser Untersuchung konnten Referenzbereiche für die einzelnen Elemente im Lebergewebe bestimmt werden. Unten sind die Werte für die drei untersuchten Gruppen in mg/kg zusammengefasst.

Weniger anzeigenDie Transfusionsmedizin stellt einen wichtigen Teil der Intensiv- und Notfallmedizin dar. Die fortbestehende Entwicklung neuer Transfusionstechniken sowie die Verbesserung der Kompatibilitätstestung trägt dazu bei, die Transfusionsmedizin möglichst sicher zu gestalten und Transfusionsreaktionen weitgehend zu vermeiden. Diese Dissertation beschreibt die Durchführung von Kreuzproben als Bestandteil der Kompatibilitätstestung beim Hund.
Im ersten Teil dieser Arbeit wurden Kreuzproben mittels des Röhrchen-Agglutinationsverfahrens seriell, sowohl vor als auch in den ersten Tagen direkt nach einer Transfusion, durchgeführt. Ziel war es zu untersuchen, ab welchem Zeitpunkt beim Hund eine Reaktion in der Kreuzprobe als möglichen Hinweis auf eine Alloimmunisierung infolge einer Transfusion nachweisbar ist. Bei 16 Hunden wurde innerhalb von 1 bis 13 Tagen (Median Tag 4) eine schwach positive Kreuzprobe gegen das Blut ihres ursprünglichen Spenders nachgewiesen. Interessanterweise wurde bei 12 Hunden bereits innerhalb von vier Tagen nach einer initialen Transfusion eine schwach positive Kreuzprobe festgestellt. Dies könnte auf eine frühe Alloimmunisierung infolge der Transfusion hinweisen, allerdings ist eine unspezifische Agglutination nicht auszuschließen.
Transfusionsreaktionen traten in diesem Teil der Studie nicht auf. Wie jedoch bereits in mehreren Studien vermutet, könnte es durch die Bildung von Alloantikörpern gegen Blutgruppenantigene mit einer niedrigen Antigenität zu einer verkürzten Lebensspanne der transfundierten Erythrozyten oder auch bei weiteren Transfusionen zu akuten Komplikationen kommen. Aus diesem Grund wird empfohlen, Kreuzproben, wenn es der klinische Rahmen ermöglicht, routinemäßig zur Kompatibilitätstestung vor jeder Zweittransfusion durchzuführen.
Im zweiten Teil dieser Dissertation wurden verschiedene kommerziell erhältliche Kreuzproben-Test-Kits im Vergleich zum Röhrchen-Agglutinationsverfahren bei verschiedenen Agglutinationsgraden auf Übereinstimmung der Ergebnisse sowie auf Praxistauglichkeit untersucht. Im Rahmen einer Pilotstudie wurden ein Gel-Röhrchen Verfahren [GR] von Rapid-Vet und ein durch Antiglobuline verstärktes Gel-Röhrchen Verfahren [AGR] sowie ein immunchromatographisches Verfahren [ICS] von Alvedia an 28 verschiedenen Empfänger-Spender-Paaren evaluiert. Im Vergleich zum Röhrchen-Agglutinationsverfahren war die Durchführung der verschiedenen Test-Kits einfacher und nahm durch eine kürzere Inkubationszeit weniger Zeit in Anspruch. Die Evaluierung ergab insgesamt eine 85,7%ige Übereinstimmung der AGR-, 60,7%ige Übereinstimmung der GR- und 35,7%ige Übereinstimmung der ICS-Methode mit dem Röhrchen-Agglutinationsverfahren. Bei alleiniger Betrachtung der makroskopisch positiven Testergebnisse war die Übereinstimmung zwischen dem Röhrchen-Agglutinationsverfahren und den Test-Kits entsprechend 100%, 62,5% und 0%. Die klinische Relevanz schwach positiver Kreuzproben ist bisher noch unklar, jedoch kann die Transfusion inkompatibler Blutprodukte zur Auslösung hämolytischer Transfusionsreaktionen führen. Sowohl das Gel-Röhrchen Verfahren als auch das immunchromatographische Verfahren zeigten daher eine ungenügende Sensitivität. Mit dem durch Antiglobuline verstärkten Gel-Röhrchen Verfahren wurden lediglich 4/9 schwach positive Kreuzproben, bei welchen mit dem Röhrchen-Agglutinationsverfahren eine mikroskopische Agglutination mit einem Agglutinationsgrad von 1+ - 2+ sichtbar wurde, als kompatibel gewertet. Bei allen weiteren Kreuzproben mit einem stärkeren Agglutinationsgrad stimmten die Ergebnisse mit denen des Röhrchen-Agglutinationsverfahrens überein. Die Durchführung hingegen war durch Entfall des Waschschrittes und eine kürzere Inkubationszeit vereinfacht und nahm im Vergleich zum Röhrchen-Agglutinationsverfahren deutlich weniger Zeit in Anspruch (Median 13,5 Minuten weniger). Aus diesem Grund bietet dieses Verfahren besonders in Notfall-Situationen eine gute Alternative zur Labormethode.

This is why microscopes were invented, to enable the observation of smaller objects through a specific arrangement of optical components, as well as the investigation of biological processes regarding disease or treatment. Through the microscope’s eye, the camera, images of various tissue stainings can be digitalized and stored on computers. At this time, image interpretation is often performed by the investigator. Manual annotation or counting is a subjective process by the observer, time consuming and therefore perhaps biased. Additional physical limitations increase the chance of misinterpretation regarding aberrations or other distortion artifacts, as well as complexity, which increases by the growing number of available parameters from the same location, notably in spatial gene expression detection. Bioimage analysis, as a newly emerging field in recent years, combines data analysis and imaging methods related to the study of biological processes and has led me to this work, where image data from various microscopy techniques were used to create supervised- and unsupervised bioimage analysis workflows validating biological questions in tissue samples from mouse and human, which were conceptually developed on the MELC system. Thereby, the MELC-system was improved to its overall image quality regarding resolution (laterally 60%, axial 250%), duration of MELCexperiments (reduction of heat and related drying of tissue sample, increase of detectable fluorescence signals) and signal evaluation (artifact removal). The application of system related artifact removal algorithms, such as image registration, illumination correction, image projections or destriping were part of required image pre-processing, which standardized subsequent image segmentation in ilastik and CellProfiler. Classification of found objects or cells benefited from clustering or dimensionality reduction methods simplifying complexity of multi parameter data sets. The application of neighborhood analysis supported the investigation with regard to spatial localizations and interaction. Based on quantitative image analysis workflows developed, including image pre-processing, image segmentation, object identification, classification and representation of the results, a general path from image acquisition to meaningful data evaluation as tool for users could was realized in case of LSM, MELC and LSFM data. Furthermore, the application of MELC and ST linked the information obtained between the protein and transcriptional level. In this way several observations were made, which helped to answer questions in immunology and neuroscience. Segmented LSM images of microglia from aging mice revealed the morphological changes and loss of processes during the course of dementia, using DFT descriptors as a compact way to describe complex shapes. The distribution of the complex compartment of stromal markers in the bone marrow of mice was heterogeneous, which can provide insights into their roles in hematopoiesis and immune cell development. Supervised and unsupervised image analysis workflows identified rare ILC populations in MELC images of mouse tonsils. In addition, their location around vessels and fibronectin fibers was detected by neighborhood analysis. Finally, SARS-CoV-2-induced tissue remodeling was observed in human lung samples from MELC and ST experiments.
Weniger anzeigenNon-tuberculous mycobacteria (NTM) are increasingly recognized for their impact on human health, causing infections, especially in individuals with compromised immune systems or underlying lung conditions. Unlike the well-known Mycobacterium tuberculosis (MTB), NTM's intrinsic resistance to common antibiotics poses treatment challenges. Diagnosing NTM infections is complicated due to nonspecific symptoms, often confused with other diseases. Understanding NTM biology is crucial for improved diagnostics and treatments. The adoption of CRISPRi (Clustered Regularly Interspaced Short Palindromic Repeats interference) and CRISPR/Cas9 in NTM represents a significant breakthrough for advancing NTM research and treatment.
The study applied the CRISPRi/dCas9 system to NTM species (M. smegmatis, M. abscessus, and M. avium), using a reporter eGFP gene for functional analysis. CRISPRi led to significant downregulation of eGFP in M. smegmatis and M. abscessus. In M. avium, reduced fluorescence, even without sgRNA, suggested the need for further optimization. CRISPRi proved effective for gene knockdown in M. abscessus.
CRISPR/Cas9 was employed to knockout the eGFP gene, resulting in loss of fluorescence in M. smegmatis and M. avium but not in M. abscessus. Assessing the system on the mps1 gene in M. abscessus revealed defects in Glycopeptidolipid synthesis, causing morphotype transition. Evaluation on endogenous genes (porin, ppiA, erm-41, and whiB7) showed successful knockout for porin and ppiA. However, targeting erm-41 and whiB7 indicated the need for system optimization.
The successful establishment of CRISPRi and CRISPR/Cas9 in NTM signifies a milestone, offering new avenues for research and potential therapies. These technologies enable precise genomic manipulation, enhancing our understanding of NTM biology and facilitating the development of more effective NTM infection treatments.
Weniger anzeigenHintergrund: Bei Patient*innen mit onkologischen Erkrankungen steigt die Nachfrage für komplementärmedizinische Verfahren. Hierzu gehören äußere Anwendungen wie beispielsweise wärmende Schafgarbe-Leberkompressen, welche in naturheilkundlichen Konzepten häufig zusätzlich während onkologischer Therapien angewandt werden. Patient*innen mit onkologischen Erkrankungen leiden häufig unter Tumor-assoziierter Fatigue (TF). Diese wird mit einer Dysregulation des Autonomen Nervensystems (ANS) und Veränderungen der Herzratenvariabilität (HRV) in Zusammenhang gebracht.
Ziel: Untersuchung der Auswirkung der äußeren Anwendung von Schafgarbe-Leberkompressen auf die Aktivität des ANS mittels HRV-Analyse bei Patient*innen mit Tumor-assoziierter Fatigue bei metastasierter Tumorerkrankung, welche eine zweiwöchige Bestrahlungstherapie von ossären oder zerebralen Metastasen erhalten.
Methoden: In einer randomisierten Pilotstudie wurden Patient*innen mit Tumor-assoziierter Fatigue bei metastasierter Tumorerkrankung, welche im Rahmen ihrer onkologischen Therapie eine stationäre Bestrahlung von entweder ossären oder zerebralen Metastasen erhielten, in zwei Gruppen randomisiert: Eine Interventionsgruppe mit wiederholten Anwendungen von Schafgarbe-Leberkompressen und eine Kontrollgruppe ohne diese Anwendungen. Mittels 24-h-EKG wurden zu Beginn (T1) und zum Ende (T2) des zweiwöchigen Studienzeitraumes Daten für die Analyse der HRV erhoben und jeweils eine Tages- (d) und eine Nachtsequenz (n) extrahiert. Anschließend wurden die HRV-Parameter des Zeit- und Frequenzbereichs sowie der Nichtlinearen Dynamik analysiert.
Ergebnisse: Im Zeitraum von September 2017 bis August 2019 erfolgte die Randomisierung von 39 Patient*innen. Die Daten von 20 Patient*innen (10 Patient*innen pro Gruppe) gingen in die Auswertung ein. Zwischen den beiden Tagesmesssequenzen im Vergleich über die Zeit (T1dT2d) konnte in beiden Gruppen ein signifikanter Unterschied der HRV-Parameter beobachtet werden. In der Interventionsgruppe nahmen die HRV und die Komplexität der Regulation der Herzfrequenz ab, als Hinweis auf eine erhöhte Aktivität des Sympathischen Nervensystems (SNS). In der Kontrollgruppe hingegen nahmen die HRV und die Komplexität der Regulation der Herzfrequenz zu, als Hinweis auf eine erhöhte Aktivität des Parasympathischen Nervensystems (PNS). Im direkten Vergleich der beiden Gruppen zeigte sich kein signifikanter Unterschied in den HRV-Parametern, mit Ausnahme des zweiten Tagesmesszeitpunktes (T2d).
Zusammenfassung: In der Interventionsgruppe mit täglicher Anwendung von Schafgarbe-Leberkompressen wurde tagsüber eine Zunahme der sympathischen Aktivität beobachtet, in der Kontrollgruppe ohne äußere Anwendungen hingegen trat tagsüber eine Zunahme der parasympathischen Aktivität auf. Aufgrund von großen Schwierigkeiten bei der Rekrutierung und Durchführung wurde die Studie vorzeitig beendet, dennoch lässt das Pilotergebnis einige Hypothesen zu. Eine Aktivitätssteigerung des SNS tagsüber könnte beispielsweise eine Verbesserung der Mobilisierung von Energieressourcen darstellen, durch welche die Patient*innen mit TF bei metastasierter Tumorerkrankung profitieren könnten. Insgesamt sind allerdings keine klaren Aussagen möglich und es gilt die aufgestellten Hypothesen in weiteren Studien zu überprüfen.
Weniger anzeigenThe present dissertation aims at systematically investigating manifestations of experience-dependent plasticity in the auditory domain, resulting from intensive musical training, utilizing analytical tools from network neuroscience. The dissertation is based on data acquired in the course of a longitudinal study investigating structural and functional changes in the auditory domain due to music training. A group of aspiring professional musicians, attending preparatory courses for entrance exams at universities of arts, and a group of amateur musicians, actively practicing in their everyday life, completed up to 5 behavioral and neuroimaging assessments in the course of one year. The dissertation consists of three studies addressing cross-sectional and longitudinal aspects of functional plastic differences and changes, respectively, ranging from a specific auditory process over unconstrained music listening to longitudinal changes in functional organization.
Weniger anzeigenHintergrund: Verbitterung ist eine komplexe Emotion und beinhaltet u.a. Frustrationen bis hin zu Aggressionen mit Rachegedanken. Der Zusammenhang zwischen Aggressionsphantasien im Rahmen von Verbitterungszuständen wird in der Forschung erwähnt, jedoch gibt es kaum empirische Forschungsergebnisse dazu, weshalb die vorliegende Untersuchung Neuland betritt.
Methode: Therapeuten*innen, die bei der Aufnahmeuntersuchung in einer psychosomatischen Rehabilitationsklinik bei Patienten*innen Hinweise auf klinisch relevante Verbitterungszustände bzw. aggressive und/oder suizidale Phantasien wahrnahmen, führten mit dem Patienten bzw. der Patientin ein vertieftes Interview bezüglich Aggressionsphantasien durch. Zu dokumentieren waren die Art und Intensität der Aggressionsphantasien sowie das auslösende Ereignis (persönliche Herabwürdigung, öffentliche Demütigung, Vertrauensbruch, Tod/Verlust). Das vertiefte Interview bildete die Grundlage für weitere therapeutische Entscheidungen. Die Patienten*innen füllten den PTED-Selbstbeurteilungsfragebogen (Post-Traumatic-Embitterment-Disorder Selbstbeurteilung) sowie die SCL-90-R (Symptom-Checkliste-90-R) aus.
Ergebnisse: Im Erhebungszeitraum wurden 3300 Patienten*innen in der Abteilung für Verhaltensmedizin behandelt. 127 von den Patienten*innen (3,84%) berichteten über ausgeprägte Verbitterungsgefühle und/oder Aggressions- sowie Suizidphantasien. Davon gaben 83,5% aggressive Phantasien an. Es fand sich ein signifikanter Zusammenhang zwischen der Verbitterungsintensität und den Aggressionsgedanken (r=0,25; p=0,017). Als häufigstes auslösendes Lebensereignis wurde persönliche Herabwürdigung (62,7%) genannt. Speziell Mobbing am Arbeitsplatz erwies sich als Risikofaktor für Aggressionsphantasien (χ2(3) = 44,2; p<0,001). Beim Vergleich der einzelnen Ereignisse (persönliche Herabwürdigung, öffentliche Demütigung, Vertrauensbruch) bezüglich soziodemographischer Daten, psychosomatischer Belastung und aggressiver Phantasien fand sich nur zwischen Verbitterung und öffentlicher Demütigung ein signifikanter Zusammenhang (t(101)=-2,07; p=0,041). Öffentliche Demütigung scheint somit als besonders belastend erlebt zu werden. In 94,1% der Fälle war das Aggressionsobjekt der Aggressor bzw. die Aggressorin selbst. 26,2% der Patienten*innen gaben an eine Person schädigen zu wollen und 12,6% lebensgefährdende Aktionen. Nur ein Drittel der Patienten*innen berichtete spontan über derartige Gedanken. Nach dem Urteil der Therapeuten*innen lag die relevante Umsetzungswahrscheinlichkeit der Phantasien bei 17,9%. 3,2% der Patienten*innen gaben Phantasien eines erweiterten Suizids an.
Diskussion: Die Daten bestätigen, dass Verbitterungsgefühle sowie Mobbingerleben im relevanten Umfang mit aggressiven Phantasien assoziiert sind und die dabei erlebte Ungerechtigkeit von besonderer Bedeutung ist. Die Art der Phantasien ist durchaus ernst zu nehmen, da bei bestimmten Rahmenbedingungen ein Ausagieren nicht ausgeschlossen werden kann.
Weniger anzeigenObjective: The term ‘primary dystonia’ is used to describe idiopathic or genetic cases of dystonia that are isolated and do not entail pathological changes. Primary dystonia, as opposed to secondary dystonia, has long been known to be lacking of any anatomical substrate. During deep brain stimulation (DBS) trajectory planning, however, we discovered T2-hyperinstensive signal alterations (SA) in the target area, especially within young patients with dystonia. Those young patients normally should not have SA.
Methods: We studied 50 MRIs from patients with primary dystonia who were implanted with DBS. An evaluation of SA volumes and total basal ganglia volumes took place, followed by 50 age-matched controls.
Results: The dystonia group has a 10-fold prevalence of SA inside the globus pallidus (GP). The biggest difference was observed in the age group that was younger than 25- year-old. A total basal ganglia volume variation was observed in the dystonia group, with larger GP and significantly smaller putamen and caudate.
Conclusions: We observed differences in the basal ganglia anatomy between primary dystonia patients and the control group. Decreases in putamen and caudate volume may indicate functional degeneration, meanwhile a bigger volume of putamen and caudate might imply overactivity.
